Mechanism of Action

Leuprolide acts as a GnRH receptor superagonist. Upon initial administration, it stimulates the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), leading to a transient increase in gonadal steroids. However, continuous exposure results in the downregulation and desensitization of pituitary GnRH receptors, ultimately suppressing the secretion of LH and FSH and reducing serum testosterone or estrogen to castrate levels.
