Overview

CJC-1295 is a synthetic growth hormone-releasing hormone (GHRH) analog developed by ConjuChem Biotechnologies. It features a modified sequence of the first 29 amino acids of GHRH with the addition of a Drug Affinity Complex (DAC) that binds to serum albumin, dramatically extending its half-life. CJC-1295 with DAC has a half-life of approximately 8 days, enabling sustained GH elevation with once-weekly dosing. A variant without DAC (CJC-1295 without DAC, also called Modified GRF 1-29) has a shorter half-life of approximately 30 minutes.

Mechanism of Action

CJC-1295 acts on GHRH receptors in the anterior pituitary, stimulating the synthesis and pulsatile release of growth hormone. The DAC modification (a lysine-linked maleimidopropionic acid group) covalently binds to circulating albumin, extending the half-life from minutes to days. This sustained elevation of GHRH analog levels produces continuous GH stimulation while maintaining the natural pulsatile pattern of GH release. The pulsatile pattern is preserved because somatostatin's inhibitory feedback remains functional, preventing GH level elevation during somatostatin-active periods.

Research Highlights

Clinical studies have demonstrated that CJC-1295 with DAC produces sustained elevations in GH and IGF-1 levels for up to a week following a single administration. Research showed mean IGF-1 increases of 2- to 3-fold lasting 7+ days. Studies explored its potential in growth hormone deficiency, lipodystrophy, and as a research tool for sustained GH axis stimulation. The without-DAC variant (Modified GRF 1-29) is used in research requiring physiological GH pulsing patterns, often combined with GHRPs for synergistic effects.

Dosage & Administration

In research, CJC-1295 with DAC has been administered at 1-2 mg subcutaneously once or twice weekly. CJC-1295 without DAC (Modified GRF 1-29) is typically administered at 100-300 mcg subcutaneously 1-3 times daily. For research use only (RUO).

Safety Profile

Phase I clinical trials reported favorable safety profiles, with mild injection site reactions being the most common adverse effect. No significant effects on cortisol, prolactin, LH, FSH, or TSH were observed. Long-term safety data is limited. Development was discontinued for commercial, not safety, reasons. Not approved by the FDA for any indication.
