Tamoxifen Citrate
Triphenylethylene SERM; tissue-selective estrogen antagonist/agonist
Mechanism of Action
Tamoxifen is a selective estrogen receptor modulator (SERM) of the triphenylethylene class. It functions as a competitive antagonist at estrogen receptors (ER) in breast tissue, inhibiting estradiol-mediated proliferation. Paradoxically, it acts as a partial agonist in bone and endometrial tissue, maintaining bone density while posing endometrial proliferation risk. Tamoxifen is metabolized by CYP2D6 and CYP3A4 to active metabolites including endoxifen and 4-hydroxytamoxifen, which have 30-100x greater ER binding affinity than the parent compound. In research contexts, tamoxifen is studied for estrogen-mediated gynecomastia prevention and HPTA recovery.

(6)Clomiphene Citrate: Evidence-Based Research Review
Clomiphene citrate (CC) is a non-steroidal triphenylethylene derivative classified as a Selective Estrogen Receptor Modulator (SERM). Originally developed for the induction of ovulation in women with anovulatory infertility, it has become a cornerstone of reproductive medicine. CC operates as a race
Tamoxifen Citrate: Evidence-Based Research Review
Tamoxifen citrate is a non-steroidal triphenylethylene derivative and the prototypic Selective Estrogen Receptor Modulator (SERM). Originally developed as a contraceptive, it has become a cornerstone in the treatment and prophylaxis of estrogen receptor-positive (ER+) breast cancer. Tamoxifen exhibi
Raloxifene: Evidence-Based Research Review
Raloxifene is a second-generation selective estrogen receptor modulator (SERM) of the benzothiophene class. Developed to address the limitations of first-generation SERMs like tamoxifen, raloxifene exhibits distinct tissue-specific effects: it acts as an estrogen receptor (ER) agonist in bone and li
Raloxifene vs Tamoxifen: Evidence-Based Comparison
Selective Estrogen Receptor Modulators (SERMs) are a class of compounds characterized by their ability to act as estrogen receptor (ER) agonists or antagonists in a tissue-specific manner. Tamoxifen, a first-generation triphenylethylene SERM, and Raloxifene, a second-generation benzothiophene SERM,
Tamoxifen vs Clomiphene: Evidence-Based Comparison
Tamoxifen Citrate and Clomiphene Citrate are triphenylethylene-derivative Selective Estrogen Receptor Modulators (SERMs) that have been used extensively in clinical practice for over four decades. While both compounds share a similar chemical backbone, they exhibit distinct tissue-specific activitie
Drostanolone Propionate: Evidence-Based Research Review
Drostanolone propionate (2α-methyl-5α-androstan-17β-ol-3-one propionate) is a synthetic anabolic-androgenic steroid (AAS) derived from dihydrotestosterone (DHT). Originally developed for the treatment of advanced breast cancer in postmenopausal women, it is characterized by its inability to aromatiz
