Toremifene Citrate
Triphenylethylene SERM; tamoxifen analog with reduced endometrial risk
Mechanism of Action
Toremifene is a chlorinated triphenylethylene SERM, structurally similar to tamoxifen (chlorine substituent at the ethyl side chain). Like tamoxifen, it acts as an ER antagonist in breast tissue and a partial agonist in bone and endometrium. The chlorine modification results in less DNA adduct formation and potentially reduced endometrial proliferation risk compared to tamoxifen. Toremifene is metabolized by CYP3A4 (not CYP2D6 like tamoxifen), avoiding the CYP2D6 polymorphism issue that affects tamoxifen efficacy. Studied for breast cancer and as an alternative to tamoxifen with more favorable metabolic profile.

(4)Clomiphene Citrate: Evidence-Based Research Review
Clomiphene citrate (CC) is a non-steroidal triphenylethylene derivative classified as a Selective Estrogen Receptor Modulator (SERM). Originally developed for the induction of ovulation in women with anovulatory infertility, it has become a cornerstone of reproductive medicine. CC operates as a race
Toremifene Citrate: Evidence-Based Research Review
Toremifene citrate is a chlorinated triphenylethylene derivative and a second-generation selective estrogen receptor modulator (SERM). Structurally analogous to tamoxifen, toremifene was developed to maintain the therapeutic efficacy of ER-antagonism in breast tissue while potentially improving the
Raloxifene: Evidence-Based Research Review
Raloxifene is a second-generation selective estrogen receptor modulator (SERM) of the benzothiophene class. Developed to address the limitations of first-generation SERMs like tamoxifen, raloxifene exhibits distinct tissue-specific effects: it acts as an estrogen receptor (ER) agonist in bone and li
Drostanolone Propionate: Evidence-Based Research Review
Drostanolone propionate (2α-methyl-5α-androstan-17β-ol-3-one propionate) is a synthetic anabolic-androgenic steroid (AAS) derived from dihydrotestosterone (DHT). Originally developed for the treatment of advanced breast cancer in postmenopausal women, it is characterized by its inability to aromatiz
