Adalank
Adamantane-modified anxiolytic peptide with enhanced membrane permeability and prolonged half-life
Mechanism of Action
Adalank is a structural analog of Selank with an adamantane moiety substituted at its N-terminus. This adamantylation significantly increases lipophilicity, enabling superior blood-brain barrier penetration compared to native Selank. Like Selank, Adalank modulates GABAergic neurotransmission and inhibits enkephalin-degrading enzymes, effectively prolonging endogenous opioid activity. The adamantane scaffold also confers enhanced metabolic stability, extending the compound's biological half-life and reducing the frequency of dosing required to maintain receptor engagement. Research suggests Adalank may modulate BDNF expression and reduce pro-inflammatory cytokines (IL-6, TNF-α) in neuronal tissue models.

Adalank and the Selank Peptide Class: Anxiolytic Peptide Pharmacology and Translational Rationale
Adalank is a synthetic analog of the anxiolytic peptide Selank incorporating an adamantane moiety. This review outlines the pharmacological rationale and preclinical evidence from the Selank peptide class that informs its development.
Overview
Adalank is a next-generation derivative of Selank featuring an adamantane group covalently attached to its N-terminus. Developed as part of a broader research program into peptide half-life extension, Adalank retains the anxiolytic and nootropic profile of Selank while exhibiting significantly enhanced pharmacokinetic properties. Preclinical in vitro data suggests improved CNS bioavailability and sustained receptor engagement.
Mechanism of Action
The core anxiolytic action of Adalank mirrors that of Selank: modulation of GABAergic signaling and inhibition of enkephalin-degrading enzymes (enkephalinase), which prolongs the endogenous anxiolytic signal. The adamantane scaffold reduces susceptibility to peptidase degradation, extending the compound's effective duration. Research also implicates BDNF modulation and anti-inflammatory cytokine regulation as secondary mechanisms relevant to neuroprotection.
Comparative Profile vs. Selank
Compared to Selank, Adalank demonstrates a substantially longer estimated half-life due to metabolic stabilization by the adamantane group. Blood-brain barrier penetration is predicted to be superior due to increased lipophilicity. The anxiolytic ceiling appears similar, though the extended duration profile may reduce dosing frequency. Unlike classical benzodiazepines, neither Selank nor Adalank appears to produce tolerance, sedation, or physical dependence in preclinical models.
Research Status
Adalank remains an early-stage research compound. While its pharmacological rationale is well-grounded in the established Selank literature, independent clinical data is limited. Most evidence is extrapolated from Selank human trials and Adalank in vitro models. Not approved for human use in any jurisdiction.
