Adalank is a synthetic analog of the anxiolytic peptide Selank incorporating an adamantane moiety. This review outlines the pharmacological rationale and preclinical evidence from the Selank peptide class that informs its development.

Overview

Adalank is a synthetic peptide analog of Selank (a heptapeptide: Thr-Lys-Pro-Arg-Pro-Gly-Pro) modified with an adamantane scaffold. Selank itself was developed as an anxiolytic and cognitive-enhancing peptide and has been used clinically in Russia. Adalank represents a next-generation analog designed to enhance pharmacokinetic properties, particularly CNS penetration and duration of action, through the lipophilic adamantane moiety.

Pharmacological Rationale

Parent Peptide: Selank

Selank has been characterized in preclinical studies as a modulator of central nervous system activity with anxiolytic effects that do not produce the sedation or dependence associated with benzodiazepines. Its mechanism is understood to involve modulation of the GABAergic system (without direct receptor binding), regulation of enkephalin degradation, and upregulation of brain-derived neurotrophic factor (BDNF). Selank has also been reported to influence pro-inflammatory cytokine signaling, including IL-6 modulation, suggesting a neuroimmune component to its effects.

Adamantane Modification

The adamantane (tricyclic cage hydrocarbon) moiety is employed in medicinal chemistry to enhance lipophilicity, metabolic stability, and blood-brain barrier penetration. In the context of Adalank, this modification is hypothesized to extend the systemic half-life of the parent Selank sequence (estimated at 2–4× longer) and improve CNS bioavailability, potentially allowing for lower dosing frequency.

Translational Evidence from the Selank Class

Anxiolytic and Cognitive Effects

In vivo studies of Selank in rodent models have demonstrated anxiolytic activity in elevated plus-maze and conflict-based behavioral paradigms, as well as cognitive-enhancing effects in contextual fear conditioning and avoidance tasks. These effects were observed without motor impairment or sedation.

Neurochemical Effects

Selank administration has been associated with modulation of monoamine neurotransmitter levels (serotonin, dopamine) and upregulation of BDNF expression in the hippocampus, consistent with both anxiolytic and neuroprotective profiles.

Immune-Neuroendocrine Interaction

Studies have reported that Selank influences the expression of inflammatory cytokines and may normalize stress-induced immune alterations, suggesting a neuroimmune mechanism relevant to stress-related anxiety.

Evidence Assessment

Adalank is a newer analog with limited directly published clinical literature. Its development rationale is grounded in the preclinical evidence base for the Selank peptide class, supported by established medicinal chemistry principles of adamantane modification. Direct clinical evidence specific to Adalank remains to be established. The evidence tier is preclinical, and the compound should be regarded as a rational analog design requiring independent validation of its pharmacokinetic and pharmacological claims.
