Overview

Epitalon (Epithalon) is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) derived from the pineal gland peptide extract epithalamin. Developed by Professor Vladimir Khavinson and colleagues at the St. Petersburg Institute of Bioregulation and Gerontology, Epitalon has been studied extensively in Russian gerontology research for its potential role in telomere biology and pineal gland function. It is one of the most recognized peptides in anti-aging research, with a body of preclinical and limited clinical literature spanning over four decades.

Mechanism of Action

Epitalon's primary proposed mechanism involves the activation of telomerase, the ribonucleoprotein enzyme complex responsible for adding telomeric repeats to chromosome ends. By upregulating telomerase reverse transcriptase (TERT) expression, Epitalon may slow the progressive shortening of telomeres that occurs with each cell division — a process strongly correlated with cellular senescence and organismal aging. Additionally, Epitalon has been shown to modulate pineal gland function, influencing the production and secretion of melatonin and other pineal peptides, which may contribute to its observed effects on circadian rhythm regulation and neuroendocrine function.

Research Highlights

In animal studies, Epitalon administration has been associated with extended lifespan in mice (12-25% increase in some studies), improved markers of immune function, and restoration of melatonin rhythms in aged animals. Khavinson's clinical studies in elderly human populations reported improvements in cardiovascular function, immune parameters, and subjective quality of life measures. A key 2003 study demonstrated that Epitalon increased telomere length in human somatic cells in vitro. Research has also explored its potential in modulating oxidative stress markers and supporting neuroendocrine homeostasis in aging models.

Dosage & Administration

In published research protocols, Epitalon has been administered via subcutaneous injection at doses ranging from 0.1 to 10 mg per day, typically over courses of 10-20 days. Some Russian clinical protocols used courses repeated 1-2 times per year. Intranasal formulations have also been explored. All dosing data derives from preclinical and limited clinical research — no standardized human dosing guidelines exist outside of investigational contexts. For research use only (RUO).

Safety Profile

Available preclinical and limited clinical data suggest a favorable safety profile, with no significant adverse events reported in short-term studies. Long-term safety data in humans remains limited. As with all investigational peptides, contraindications, drug interactions, and long-term effects have not been fully characterized. Not approved by the FDA for any indication in the United States.

FDA 2026 Regulatory Update

As of April–June 2026, this compound was among the 14 peptides formally removed from the FDA's Category 2 restricted compounding list following review by the Pharmacy Compounding Advisory Committee (PCAC). This regulatory shift permits licensed compounding pharmacies to produce and dispense this compound under appropriate oversight, subject to final 503A Bulks List adjudication expected following the July 23–24, 2026 PCAC meeting. This does not constitute FDA approval for therapeutic use. All information on this page is for research purposes only.
