Telomerase Activation and Cellular Senescence Pathways

Overview

Telomere shortening is a hallmark of cellular aging. Research into telomerase-activating compounds, mitochondrial-derived peptides (MDPs), and senescence-pathway modulators provides insights into aging biology and longevity mechanisms.

Telomere and Telomerase Biology

Telomerase is a reverse transcriptase maintaining telomere length in stem and germ cells. Synthetic peptides studied for telomerase modulation include:

  • •Epithalon (tetrapeptide AEDG): Pineal-derived, studied for telomerase activation
  • •Thymosins: Thymic peptides with immunomodulatory properties
  • •GDF11 and related myostatin antagonists

Mitochondrial-Derived Peptides (MDPs)

MDPs (MOTS-c, SS-31, humanin) are endogenous peptides encoded by mitochondrial DNA with roles in:

  • •Oxidative phosphorylation efficiency
  • •Mitochondrial stress response
  • •NAD+ metabolism
  • •Senescence resistance

Preclinical Evidence

Rodent lifespan and healthspan studies document extended longevity with certain MDP interventions. Cellular senescence models show enhanced autophagy and reduced p16/p21 markers with telomerase-modulating compounds.

Research Significance

These compounds serve as tools for understanding cellular aging mechanisms and longevity biology in laboratory research.
